Isolation of an Acidophilic Cellulolytic Bacterial Strain and Its Cellulase Production Characteristics

The aim of the study was to isolate and identify a highly efficient cellulolytic bacteria strain that can be used in acidic environments, then investigate its cellulase production characteristics for effective utilization agricultural waste. For this purpose, we set series isolation screening steps, 21 strains were isolated from soil, an acidophilic labeled as B13-2 with high screened using Gram’ iodine method activity assay; it identified Raoultella terrigena. Lastly, culture conditions such incubation time, temperature, pH, carbon sources, nitrogen inoculum size optimized via single-factor experiments, on basis, response surface methodology optimization goal. results showed optimum time is 3.1 days, temperature 29.9 °C, pH 4.1, 1.50%, reached maximum 13.503 U/mL, which about 140% higher than before optimization.
